Valz N is a combination type drug that has an antihypertensive effect on the body. It includes an angiotensin II receptor blocker and a thiazide diuretic.

Substance valsartan is a peripheral vasodilator, it acts on the body as a hypotensive agent and diuretic. Valsartan is a specific blocker of AT1 angiotensin II receptors; it does not inhibit ACE. The substance has no effect on the level of cholesterolglucose TG, uric acid. With a decrease in pressure in humans, there is no change in heart rate.

Substance hydrochlorothiazide is a medium power diuretic. Compared with furosemide, its diuretic effect is weaker. Under the influence of this substance, the excretion of potassium in the urine increases, as well as the excretion of bicarbonates and phosphates.

Hydrochlorothiazide increases excretion magnesium, in the body under its influence are delayed calcium ions and urata. Arterial pressure decreases due to a decrease in bcc, a change in the reactivity of the vessel wall, an increase in the level of depressive effects on the ganglia and a decrease in the decrease in the pressor effect of substances with vasoconstrictor properties.

Pharmacodynamics and pharmacokinetics

The simultaneous use of hydrochlorothiazide and valsartan reduces systemic bioavailability by approximately 30%. This interaction does not affect the overall effectiveness of the use of substances in combination.

Valsartan is rapidly absorbed, absorbed variable. Its bioavailability is 23%. The highest concentration of a substance is observed two hours after the use of drugs. Valsartan binds to blood proteins by 94-97%. Its half-life is 9 hours. Most of it is excreted unchanged, 70% through the intestines, 30% through the kidneys.

Hydrochlorothiazide after oral administration it is absorbed quickly, its bioavailability is 60-80%. The highest concentration of the substance is achieved after 2-5 hours. With plasma proteins hydrochlorothiazide binds to 60-80%. Its half-life is from 6 to 15 hours. It is excreted through the kidneys, while more than 95% comes out unchanged.

The greatest hypotensive effect in the treatment with the drug is observed in the first two to four weeks of therapy.

Valz N lowers excretion quinidine.

With simultaneous treatment, the effectiveness of hypoglycemic oral drugs, anti-gout drugs, epinephrine, norepinephrine.

The frequency of development of allergic manifestations to allopurinol.

Valz N reduces the level of excretion of cytotoxic drugs through the kidneys and increases their myelosuppressive effect.

Drugs that intensively bind to blood proteins in the body (clofibrate, indirect anticoagulants, NSAIDs), increase the diuretic effect.
